D12 is a fully human monoclonal antibody targeting PD-1, isolated from the synthetic AMG phage display library. It binds a novel epitope on PD-1 and induces an unusual conformational change at the receptor’s N-terminus. Despite limited overlap with the natural ligand-binding site, D12 effectively blocks PD-1 interaction with PD-L1 and PD-L2 through steric hindrance. The antibody shows high-affinity binding, recognizes PD-1 on activated human T cells, and cross-reacts with Cynomolgus monkey PD-1.
